Uv-Vis Procedure
PROCEDURE
1) A standard was prepared in solvent and was placed into sample holder for the purpose of calibration curve.
2) The liquid sample was also prepared in a similar manner and placed into sample holder.
3) The UVProbe Software was opened and the method(M) was set to wavelength of 800-0.0 at medium speed for measuring liquid sample absorbance , and the method was saved in a file according to sample name.
4) The standard for baseline measurement was run. (The first run using an empty cuvette followed by the standard sample)
5) Sample measurement was continued.
6) All the results were saved and the spectrum produced was saved.
7) The main component in UV-VIS unit along with the diagram of the unit was drawn.
8) Spectrum obtained was analyzed based on wavelength peak and compared against calibration curve.
9) The analyte concentration found in the unknown was determined.
10) All the findings were summarized based on the spectrum obtained and related information derived from the experimental work and from references.
STANDARD OPERATING PROCEDURE FOR UV-VIS UNIT
Start-Up
A. Equipment
1. The power supply was connected to the unit.
2. The unit and PC was switched on.

B. Steps to Perform Analysis
1. The UVPorbe Software was opened and CONNECT button was clicked. A series of tests is automatically conducted which may take about 5 minutes. All checklist is made sure to be green.
2. Prior to analysis , the method was set according to requirement, by clicking the M button.
3. The method was then saved based on sample name and the FILE menu was clicked. The method is then saved as on a created new folder.
4. Calibration must be done using BaSO4 for solid sample or solvent for liquid (50%ethanol and 50% DI water / 70% ethanol and 30% in DI water) containing the analyte (five concentration of analyte plus bank i.e without sample) as standard calibration.
5. The BaSO4 for solid or solvent for liquid cell was placed at sample holder. The BASELINE icon
