Peroxidase Enzyme Lab Report
In this lab the peroxidase enzyme is tested in a dormant avocado seed as well as an avocado seed undergoing the process of germination. A gas pressure will be used to test the seeds and see if the peroxidase enzyme is present in either of the seeds. A catalyst is very similar to track spikes. Spikes increase a runner’s speed, as a catalyst speeds up the chemical reaction time in a plant. Neither the catalyst nor shoes are changed in these actions. Enzymes are macromolecules that act like catalysts. A catalyst will lower the activation energy within a seed, which will allow the reaction to go quicker. An avocado seed is used in this experiment. For this type of seed to start the process of germination, the favorable climate is typically very warm. If someone was to plant an avocado seed and see the quickest results, they should place the seed in the sun. Along with waiting a few days between watering the seed, allowing the soil to dry out. For the seed to germinate properly, the bottom half of the seed will be emerged into water. A dormant avocado seed and an avocado seed in the process of germinating will each be tested, using a gas pressure. This tool will record if the peroxidase enzyme is present in either of the seeds. The seed undergoing germination will have awakened enzymes however the dormant seed will have un-hydrated enzymes. Depending on the amount of gas released from each seed will determine how much catalyst activity is going on in the seed. The independent variable is the amount of time that the seed will be tested for gas. The dependent variable is the gas pressure and how much gas is released from each seed. The peroxidase enzyme catalyzes the change form hydrogen peroxide into water and oxygen gas. It’s safe to assume that the dormant seed will not produce as much gas as the awakened seed

    Four environmental factors of enzymes were tested in lab. The changing of pH, substrate concentrations, temperature, and an inhibitor (NaCl) and the effects it hade on the enzyme turnip peroxidase. Enzymes are biological catalysts which increase reaction rates by lowering the activation energies of substrates. A substrate is a reactant that interacts with the enzyme. The enzyme and substrate can be viewed as the recently discovered "induced fit model", which suggests enzymes are flexible and dynamic things that change their shape so all these substrates (reactants) can become catalyzed when the activation energy is lowered and the reactions happen a lot faster. Sometimes in cells though it may not need a particular substrate (reactant) so an inhibitor comes into play. Inhibitors are basically regulators that inhibit (disallow) the process of catalysis to take place within a particular substrate. There are two types of inhibitors that occur in such reactions. Competitive inhibiters are remarkably similar to the substrate, so much that it can match interchangeably with the substrate, thus leading to a halt in the production of the intended product. Another variety of inhibitor is the allosteric inhibitors. Allosteric inhibitors change the shape of the enzyme by binding to a different site other the active site, which is the usual site for catalysis. Usually the allosteric inhibitors make contact with the side of the enzyme opposite the active site. In some occasions however an allosteric activator is introduced and functions by connecting to the enzyme in a way that it allows for easy access for a substrate to the active site. Allosteric activators are the opposite of allosteric inhibitors. These regulators (both inhibitive and active in function) help keep the cell in homeostasis by not allowing too much or too little of a needed or not needed product to be produced..
