Observing Bacteria and Blood

Questions:

A. List the following parts of the microscope and describe the function of each

A. Eyepiece lens B. Tube C. Revolving nosepiece/turret D. Objective lenses E. Stage F. Iris Diaphragm G. Illuminator H. Coarse Focus I. Fine Focus J. Arm K. Stage Clips

B. Define the following microscopy terms:
a. Focus: A means of moving the specimen closer or further away from the objective lens to render a sharp image.
b. Resolution: The ability of a lens system to show fine details of the object being observed.
c. Contrast: A circular plate placed on the stage of a low power microscope. One side is white, the other is black. It can be flipped around depending on the coloration of your specimen.

C. Describe your observations from fresh yogurt slide you prepared in Part III. Several cells majority Bacillus and cocci cells. D. Were there observable differences between your fresh yogurt slide and the prepared yogurt slide? If so, describe them. Only perhaps that there was far more Cocci cells then Bacillus

E. Describe the four main bacterial shapes

1. Cocci - Spherical or oval bacteria having one of several distinct arrangements based on their planes of division
2. Bacillus - Rod shaped bacteria. Bacilli all divide in one plane producing a bacillus, streptobacillus, or coccobacillus arrangement.
3. Spirillium – Rod shaped bacteria that appear thin and spiral shapped.
4. Vibros - Oblong shape that has a similar shape to peanut shells in that their curvature.

F. What are the common arrangements bacteria are found in? Cocci, Diplococci, streptococci, staphylococci, bacillus, diplobacillus, streptobacillus.

G. Were you able to identify specific bacterial morphologies on either yogurt slide? If so, which types? I believe I could see cocci, and diplobacillus.

H. Describe the cells you were able to see in the blood smear. Red blood
