ABSTRACT
Biofuels are renewable and environmentally safe and refers to a nonpetroleum based fuel obtained from the transesterification of vegetable oils or animal fats. Bioethanol and biodiesel are two important forms of biofuels. Non-edible oil obtained from the seeds of Jatropha curcas is the potential source of biodiesel. Therefore it is necessary to find out the varieties of Jatropha which contain maximum oil percentage. Tissue culture offers a reliable method to produce quality plants in large scale. Present paper highlights in vitro regeneration protocol for the species. Keywords: Biofuels, in vitro regeneration, Jatropha curcas

INTRODUCTION Due to rapidly diminishing reserves of the fossil fuels and increasing effects of pollution from these fuels, there is an urgent need of eco-friendly alternatives to fossil fuels. Many energy fuels are being investigated. The best alternative fuel of significance for the future is biofuel. Biofuels refers to a non-petroleum based fuel obtained from the transesterification of vegetable oils or animal fats. This special type of fuel is mostly produced from bio waste. The worldwide awareness regarding global warming has popularized the use of biofuel. They are highly effective in minimizing the extent of greenhouse gas emission. Owing to the limited availability of fossil fuels, the use of biofuels becomes all the more important. They are renewable and environmentally safe. Non-edible oil seeds of forest origin are a promising option for producing biofuel. Jatropha curcas, the biodiesel plant is a potential source of such non-edible oil seeds. Among the many species, which yield oil as a source

    The purpose of this experiment was to compare biodiesel to other fuels. The ecotoxicity of biodiesel was estimated, along with that of other biofuels through the germination of seeds in the presence of different concentrations of fuels. This resulted in biodiesel having an average of 79.4% germinated seeds in a 10% solution of fuel, compared to other fuels whose percentage of germinated seeds decreased with higher concentrations of fuel. In addition, biodiesel was also synthesized and its heat of combustion was found through the use of calorimetry. The heat capacity for the calorimeter was based on the heat of combustion for ethanol, which was then used to determine the heat of combustion for biodiesel. The average heat of combustion for biodiesel was ∆Hcomb= -13090.83 kJ/mole, as opposed to the theoretical value of -11496.6 kJ/mole, which resulted in a 13.9% difference. Furthermore, taking into consideration the ecotoxicity of biodiesel, its effect on the environment, and fuel efficiency, biodiesel proved to be the more favorable fuel relative to the other fuels tested.     Energy is the common currency of all living things; something all species need. While plants rely directly on the sun for energy, humans rely on an array of sources like wind, fossil-fuel, water, solar, and more recently biofuels. Of these biofuels, which are fuels obtained directly from living plant matter, ethanol and biodiesel are the most commonly used. These two sources are typically used to fuel cars, trucks, aircraft, and small-engine machines. Biofuels have garnered much attention because they are derived from renewable resources like corn and soybeans rather than non-renewable fossil fuels. The main advantage to biofuels has always been that they give off fewer green-house gasses (GHG), particularly carbon,that petroleum-based fuels are notorious for emitting. Analyzing biofuels from an environmental prospective, they seem to be a positive alternative to replacing society’s addiction to fossil-fuels. However, the amount of fuel need to produce these crops and the amount of cropland needed to supply a nation with biofuel is often ignored in the comparison. Besides requiring high input of energy, space, fertilizer, and pesticides, switching our economy to a biofuel reliant one will lead to deforestation—additional emission of GHGs—and higher priced food.     Corymbia is credited with high growth rate and multiple uses and is planted to meet the ever increasing demand for timber. The Corymbia hybrid selected for the study, has displayed about two times height, girth and collar diameter as compared to its parents. Micropropagation has been a powerful tool for the multiplication of such hybrids by maintaining their hybrid vigour. To reduce mortality and release the stress upon transplantation as well as to provide fast growth, establishment of ectomycorrhizae on micropropagated plantlets is suggested as these help in nutrients uptake and reduce environmental stresses. Therefore, the present work was undertaken with the objective of studying interaction of Laccaria laccata, an ectomycorrhizal fungus, with micropropagated plantlet of the hybrid under in-vitro conditions. Nodal segments containing axillary buds from newly developed fresh shoots were taken and treated with 1% carbendazim (50% WP) for 10 minutes followed by surface sterilization using 0.1% mercuric chloride for 2 minutes. The explants were incubated in Murashige and Skoog (MS) medium supplemented with 3% sucrose, BAP (2 mg/l) and NAA (1 mg/l) for axillary bud induction and shoot initiation. Shoot multiplication was achieved in the same medium and regular subculturing was carried out on MS with BAP (1 mg/l). After 4-6 subculturing sufficient juvenility was achieved and thereafter half strength MS medium devoid of plant growth regulators was used to get elongated shoots to induce rooting. Best rooting was observed in half strength MS medium supplemented with IBA (0.1 mg/l). Laccaria laccata was multiplied using Modified Melin Norkrans (MMN) medium and inoculated into rooted plantlets in vermiculite having MMN/ MS medium or in soil under in-vitro conditions. Plantlets died within 1 and 2 months of inoculation in vermiculite and soil respectively.
