Daratumumab Summary
In 2015, Lokhorst et al examined the effects of giving a new drug called daratumumab by itself. Daratumumab is an antibody that targets specific receptors more commonly found on malignant myeloma cells compared to normal cells. The study was a multiphase trial that covered Phases 1 and 2. Part 1 had 32 patients, and the goal was to investigate safe dose ranges of the drug. Part 2 had 72 patients, and the goal was to investigate the efficacy of the established doses from the first phase. Part 1 could not identify a maximum tolerated dose; even the highest dose had modest adverse effects. Part 2 found that patients who received a higher dose had a higher likelihood of the drug being effective and a higher likelihood of progression-free survival
