Introduction:

When conducting an experiment with microbial organisms there often is change to the bacteria itself and has gone into mutations therefore causing evolution. In the twentieth century there has been many medical breakthroughs. One of those remarkable breakthroughs was Fleming’s discovery of the antibiotic penicillin. That antibiotic had the ability to fight off bacterial diseases and infections through microbial antibacterial chemicals. Through a couple of years the bacteria grew resistant to the penicillin. That was due to gene mutation such as involving genes, which encodes proteins that are capable of deactivating or exporting the antibiotic (Liu, 2012). Over time the bacteria E. coli became resistant to the antibiotics, and once resistant then the medicine (penicillin) has no effect (Doughari 2012). The purpose of the experiment is to understand the processes by which genes can be inserted into plasmids, also to gain some experience in cloning genes via E. coli, lastly to understand the medicinal and other implications of gene transfer among organisms (Hoot, Wimpee, 2012). To test these purposes you would need to gather petri dishes, ampicillin, calcium chloride, and plasmid pJE202.
My hypothesis is that if E. coli maintains the plasmid pJE202 then colonies will resist ampicillin and be bioluminescent. My predictions with accordance to the variables to this particular lab are as such: that Plate A (no plasmid or ampicillin) will show colonies but no bioluminescence, Plate B (Plasmid DNA but no ampicillin) will show colonies and bioluminescence, Plate C (no plasmid but ampicillin present) will show no colonies and no bioluminescence, and Plate D (Plasmid DNA and ampicillin present) will show colonies and bioluminescence. For the variables in this lab are two, first would be independent and second dependent. The independent variables are plasmid DNA and the ampicillin. The Standardized variables are agar plates.     Two experiments were done to identify an unknown plasmid. The success of these experiments came from the use of modern day technology involving gel electrophoresis. First, bacterial transformation to E. Coli DH5 was performed on our unknown plasmid along with two known plasmids, pAMP and pKAN, and a negative control TE, a buffer without DNA. By performing confluency streaking of bacteria in plates containing antibiotics, we were able to examine the recombinant DNA of the bacteria. After incubation of the plates, we analyzed the samples and found that our unknown plasmid reacted positively on the LB/AMP plate. There were a total growth of three colonies on the LB/AMP plate and a negative result on the LB/KAN plate. With this data along with the positive reaction of pAMP on the LB/AMP plate, we came to the conclusion that our unknown plasmid was pAMP. In our next experiment, we analyzed the DNA via gel electrophoresis. First, we had to treat our unknown plasmid. Three treatments were performed: Uncut (U), single cut (S) with HindIII, and double cut (D) with HindIII and Bam H1. The gel was then stained with Ethidium Bromide, often used in chromatography, in order for us to view the gel under UV light. A photograph of the result was then printed out. This allowed us to determine the migration of each sample along with the number of base pairs in each fragment. Standard fragments of DNA were used to determine the size of our unknown plasmid, which at this point was pAMP. With the use of both pKAN and pAMP plasmid maps, we were able to solidify our conclusion that the unknown plasmid was pAMP.…

    Escherichia coli is a bacterium that can affect our health or even kill. Like most bacteria, E. coli is able to change and progress into different forms based on genetic changes that they can go through. One example of this genetic change is shown in the E. coli becoming immune to ampicillin is blood infections. Because ampicillin has been used so frequently to fight the symptoms of an E. coli infection, the bacteria has been able to change itself genetically by producing more of an inhibitor resistant TEM in order to continue it’s genetic line and reproduce causing infections in humans (Walters-Toews, et al. 2011). Another example from the science field would be an experiment that suggests that E. coli is not only becoming resistant to ampicillin, but also other antibiotics including Cotrimoxazole and Cefuroxime (Renal & Urology News, 2007). This experiment is meant to prove that through genetic transfer using plasmid DNA, the E. coli can become bioluminescent and immune to the ampicillin.     Salmonella is a common bacterial disease that affects the intestinal tract. It commonly lives in human and animal intestines and are shed through feces. Humans are affected by it mostly from contaminated food or water sources. The bacteria is larger than a virus but is visible to the eye with a a microscope. It is a rod-shaped, gram negative, non-motile bacteria that does not form spores. It infects the cell, multiplies from within it, then, burst the cell. People with salmonella typically develop diarrhea, fever, and abdominal cramps within 72 hours. Most healthy people generally recover within a few days without specific treatment. In some cases diarrhea causes one to become dehydrated to the point of requiring prompt medical attention. Life-threatening complications can also occur if it spreads beyond your intestines. By August 28, 2008 there were a reported 1442 cases of the rare Santipaul serotype of samonella food poisoning in 43 states throughout the United States, the District of Columbia, and Canada. The U.S. Food and Drug Administration suspected that the contaminated food product was a common ingredient found in fresh salsa such as fresh jalapeno, raw tomatoes and fresh cilantro.
