In this TLC lab, we explored the relative polarities of Acetylsalicylic Acid, Ibuprofen, Acetaminophen, and Caffeine based on their functional groups. The most overall polar analgesic is considered to be the analgesic that contained the most polar function groups. The amine and amide functional groups are the most polar. In this experiment, we found that both these functional groups are found in Caffeine. As a result, Caffeine was the most polar out of Acetylsalicylic Acid, Ibuprofen, Acetaminophen, and Caffeine. Aside from Caffeine, Acetaminophen has one amide functional group. This makes it the second most polar analgesic. In addition, Acetylsalicylic Acid has an ester group. It is important to note that acetic acid is the non-polar component of the experiment. On the other hand, the stationary phase corresponded to the Silicon on the TLC plate. From the dipole in Silicon and Oxygen bonds, this is regarded as the polar component of the experiment. Compared to the mobile phase, the stationary phase results in the polar functional groups to be a stronger attractant. As a result, the most polar functional groups will travel up less the TLC plate. Due to the non-polar compounds not sticking to the stationary phase as strongly as the polar compounds do, non-polar compounds should have a larger Rf value than polar compounds. As previously stated, the least to most polar is Ibuprofen, Acetylsalicylic Acid, Acetaminophen, and Caffeine respectively. This shows that ideally, Caffeine has the lowest Rf value while Ibuprofen should have the largest Rf value. In the Observations section, we calculated that Ibuprofen’s Rf value is .81, Acetylsalicylic Acid’s Rf value is .64, Acetaminophen’s Rf value is .46, and Caffeine’s Rf value is .10. We used this specific eluent because it is polar. Since the analgesics used in this lab contained different functional groups, some were more polar than others. So, the polar eluent with a non-polar TLC plate assists in illustrating the different polarity strengths and determining the analgesic drugs in a mixture. Furthermore, the acetic acid component of the eluent used helps remove the excessive Ibuprofen and Acetylsalicylic Acid. This is done by restraining their ionization. Overall, ethyl acetate containing 0.5% acetic acid was effective because the various analgesics in Anacin, Excedrin, Motrin, and No-Doz were all clearly separated and

    Acetaminophen is a common analgesic used to treat mild pain. We synthesized acetaminophen by reacting p-aminophenol and acetic anhydride. The amine group on the p-aminophenol reacted with the center oxygen atom on the acetic anhydride to form an amide. We reacted 0.210g of p-aminophenol with 0.240ml of acetic anhydride in the presence of heat, and then cooled the solution in an ice bath until crystals formed. The solid acetaminophen was filtered from the solution and then subjected to a recrystallization using a 50:50 water-ethanol solvent. 0.1484g of crude acetaminophen was measured and after purification 0.0669g of pure acetaminophen was collected.     The purpose of this experiment is to investigate the composition of a simulated pharmaceutical preparation Panacetin, a proposed type of pain-killer. Panacetin is typically made up of sucrose, aspirin, and acetaminophen, but the third component in this experiment is unknown. The unknown component is suspected to be a chemical relative of acetaminophen, either acetanilide or phenacetin. Using techniques such as extraction, evaporation, and filtration, the three components will be isolated based on their solubilities and acid-base properties. The percent composition of Panacetin will also be deduced based on the masses of the three dried components; this is done to verify the composition attained is consistent with those listed on the preparations label. As a result of this investigation, my teammates and I allowed the Panacetin to undergo gravity filtration and separation techniques in order to identify whether there are any discrepancies in the components of the Panacetin. Furthermore, recrystallization and purification methods were used to determine if the unknown substance were similar in properties to either of the suspected unknown substances by comparing factors such as melting points to the chemical properties of phenactin and acetanilide. The results were as expected, based on the molecular weights and ratios of each separated chemicals, as well as the boiling point of the unknown it was determined that these ranges were close enough to indicate that the label is reasonably accurate in its composition. To add on however the identity of the unknown component differed from what the label indicated. In the end, the percentage composition attained based on our observations and yield confirmed that indeed the chemical composition of Panacetin were as indicated on the preparations label. The identity of the unknown component however suggested that the preparation did not contain acetaminophen as indicated, but instead was consistent with the chemical properties…
