Practical 1
Title: Tissue Processing
Aim: To examine human cell and tissue under the microscope
Introduction:
Tissues from the body taken for analysis of diseases must be processed in the histology laboratory to produce microscopic slides that are viewed under the microscope. These tissues are usually fixed into paraffin. Microscopic analysis of cells and tissues requires the preparation of very thin, high quality sections mounted on glass slides and properly stained to demonstrate normal and abnormal structures. Tissue processing may be performed manually or with the help of automated tissue processor. This process is important so that the tissue specimen can be sectioned easily. The main steps in this process are dehydration and clearing.

The tissue is infiltrated with paraffin wax 2 for 2 hours.

6. After infiltration of tissues enclosed in cassettes, each tissue section is moved from its labeled cassette into a mold filled with molten paraffin.
7. The labeled cassette is placed atop the mold and it is placed on chilling plate to solidify the paraffin wax (block).
8. The tissue contains cassette is then detached from the mold and it is used for section cutting. (Microtomy).
Questions:
1. What are the advantages of automated tissue processor?
Automated tissue processor saves time, decreases human error, effective fluid circulation.
2. Briefly describe about clearing agents.
Clearing agents are used to remove the dehydrant from the tissue. They also make tissue transparent by raising refractive index of any specimen. Clearing agents need to miscible with both alcohol and paraffin wax.
