Introduction Scientists have recently discovered that resistance to antibiotics may not be such a new thing. Evidence of bacteria samples in Canadian permafrost proposes that these resistances have been around for at least 30,000 years (Luiggi 2011). In our required pre-lab reading, we learned tuberculosis is becoming increasingly drug-resistant, giving proof that bacteria can adapt to necessary changes in order to survive (Barry 2009). The addition of acetyltransferases, enzymes that help move acetyl groups, to bacteria can make it more resistant to antibiotics containing aminoglycosides (Norris 2010). An aminoglycoside is simply just an antibiotic whose structure consists of amino-modified sugars. These antibiotics are effective in fighting off gram-negative bacteria in patients with infections (Schussler 2012). The challenge given to the class was to test two unknown strains of E. coli, which were “potentially” resistant to an aminoglycoside antibiotic, and see how susceptible or resistant the E. coli would be to the antibiotics.
Hypothesis: Since Dr. Engin H. Serpersu added acetyltransferases to both the “unknown” strains of E. coli, only the aminoglycosides, kanamycin and amikacin, would be affected. Prediction: The zones of inhibition of kanamycin and amikacin would be smaller because of the effect that the transferases would have on them.

Methods
First every group in the class was given two agar plates. Each group streaked their two plates with a wild E. coli strain and then divided each plate into four sections. The control group, a disk with no antibiotics, was placed in one quadrant, while three types of antibiotics were placed in the other three quadrants of both plates, totaling six different antibiotics. The antibiotics we used were as follows: ciprofloxacin, tetracycline, kanamycin, amikacin, triple sulfa, and penicillin. The class received four more agar plates per group

    Bacteria growth is known to be either augmented or impeded by a number of various factors; in this experiment, our group tested how E. coli is affected by penicillin as well as how different temperatures can affect bacteria growth. We know that antibiotics are generally specialized against certain types of bacteria—more specifically, some are most effective against Gram-negative, some against Gram-positive, and some are more broad spectrum. Given that E. coli is known to be a Gram-negative bacteria, our hypothesis was that Penicillin (narrow Gram-positive spectrum) would not be effective against E. coli; we also hypothesized that increasing temperature in the environment of the bacteria would be directly increase overall growth. We tested these hypotheses by preparing five different petri dishes, each containing cultures of the bacteria E. coli. In two of these dishes (set up identically), containing plain agar with no additional nutrients, we added small amounts of Penicillin to half the plate and water to the other half. We then observed the plates after one week’s time. For the other three plates we stored one dish at 4°C, one at 22°C, and one at 37°C; no antibiotics were added to any of these three dishes. Lastly, we then compared bacterial growth in the dishes that had been stored at different temperatures. Our final results indicated that Penicillin is not effective against E. coli because it is a narrow Gram-positive antibiotic that works by inhibiting an enzyme required for bacterial cell wall formation but some E. coli produces a slightly different version of the enzyme so reproduction may be slowed but will not stop. The other aspect of our hypothesis that temperature affects growth was found to be true as demonstrated by the plates stored at higher temperatures exhibiting substantially more growth.     The fact that each bacterial species, and even some of their respective strains, responds uniquely to a given antimicrobial makes it necessary to have methods, which provide researchers and clinicians with measurable susceptibilities. This need has become prevalent since the rising of later generations of antimicrobial compounds achieved by chemical modifications of the naturally occurring antibiotics. By utilizing the Kirby-Bauer method, researchers and clinicians are able to determine the susceptibility of given microorganisms to different antibiotics and antimicrobials. While antibiotics specifically target bacteria, antimicrobials cast a wide net, nonspecifically targeting microorganisms and viruses. During this lab, we tested the effects of both antimicrobials and antibiotics on different bacterial cultures. By creating different lawns of bacteria and placing on each of them disks soaked in different antibiotics and antimicrobials, we were able to observe the susceptibility of the differing bacteria to several agents. This process that we practiced is the Kirby-Bauer method. As stated before, antibiotics and antimicrobials serve differing purposes. Antibiotics are substances, usually in the form of medication, that destroy bacteria or inhibit the growth of bacteria. On the other hand, antimicrobials act in destroying or inhibiting the growth of all microorganisms. One type of antimicrobial is an antiseptic, which acts to reduce the presence of microorganisms without harming living animal tissue (El-Mayas, 2013). It is important to note that while we were able to measure the susceptibility of these agents, we cannot conclude that resistance is present because further investigations would have to be conducted to determine if the microorganism has developed a metabolic capability to withstand the agent’s mechanisms and effects (Maxwell, 2013).…

    To study whether a microbial product qualifies as an effective antibiotic, a standard procedure called the Kirby-Bauer method is employed. This method, which is the procedure recommended by the US Food and Drug Administration, was devised by William Kirby and A.W. Bauer in 1966. In the current protocols involved in the Kirby-Bauer method, Mueller-Hinton standard agar is used as the medium for bacterial culture. The pH of the standard agar is 7.2 to 7.4 and it is poured exclusively to a depth of 4 mm. The medium is heavily inoculated with bacteria and paper disks containing enough of the antibiotic under study to create an optical density of 1 (the McFarland standard) are placed on top of the cultures. By examining the results of incubation in the form of a zone of inhibition around each disk after incubation, it can be determined how effective each antibiotic is against any given bacterium. A minimum inhibitory concentration can then be deduced for the given antibiotic vs. the specific bacterium tested so that appropriate dosage may be determined.
