Title:
Testing for the Presence of Starches, and Proteins
Purpose:
To test for the presence of starches and protein macromolecules, to introduce the concept of variations in chemical compounds.
Hypothesis:
If biuret is blue after a test for protein, then it is a negative result because biuret reagent is blue to begin with. If a solution is pinkish purple, or purple, then the test for protein is positive. The test solution for starches is yellowish brown. If any substance that is yellowish brown when mixed with iodine, then the test for starches is negative. If the solution turns purple, or a dark purplish black, then the test for starches is positive. If DI water, and sucrose are tested for proteins using biuret reagent, then they will show a negative result. To explain, both DI water, and sucrose alone do not contain a trace of protein. In addition, deionized water is commonly used in experiments as a negative control. In both experiments, if the DI water is tested for a protein or starch, then it will come back as a negative. If milk solution, and 50% egg white solution are tested for proteins using the biuret solution, then there will be positive results because both of these are composed of protein macromolecules. The use of condensed milk brings about an even stronger positive result for protein than regular refrigerated milk, since condensed milk is higher in protein. If DI water, onion juice, and sucrose are tested for starches using iodine, then there will be a negative result for starches. This is because there are not any traces of starch in these solutions. On the other hand, if 1% starch solution is tested for starch using iodine, then there will be a positive result for starch since there is starch present.

4-5 Test Tubes
Transfer Pipet
Protective goggles
Deionized Water (DI water)
Evaporated Milk
50% Egg White Solution
1% Sucrose Solution
1% Starch
Onion juice
Sucrose

The protein test’s purpose was to determine all the test materials that contained a protein by using biuret solution. The procedure for the Spit lab was to perform the starch test and reducing sugar test on both crackers. Then chew up the cracker that doesn’t have reducing sugars. Next, test the sample for a reducing sugar. The results of the Chemical Aspects lab were that starch, potato juice, cracker solution, and cereal contain starch. In the reducing sugar test glucose, fructose, lactose, and milk contain a reducing sugar. In the grease spot test, salad oil, milk, and chips are lipids. In the dye test, salad oil and chips are fats. Egg albumin, cereal, and cracker mixture show that they are proteins. In the Spit lab the saliva and cracker test showed up positive.     In my first experiment to test for proteins the egg white with the Bluret reagent should come out positive for being a protein. The pepsin in the Bluret reagent should be positive and should be a protein. The test for sugar in the Bluret reagent should be negative and I don't think it is a protein. The test with the distilled water with the Bluret reagent should be negative and is not a protein. In the second experiment to test for glucose with the Benedict reagent sugar should be positive for glucose. Starch mixed with the Benedict reagent is probably positive for glucose because it is a polysaccharide. Onion juice mixed with Benedict reagent is probably positive for glucose because it is a fructose. Distilled water with Benedict reagent should be negative for glucose. The third experiment to test for starch using with and without the IKI reagent by looking under a microscope a slice of potato cells should have several shapes to it and with the IKI reagent should look have more of a stronger shapes and color to it. The onion cells should have a more stringy look to it and with the IKI more color. The fourth experiment is testing for lipids using the Sudan III on a potato, onion, oil and dH2O. I believe the oil will determine it is a lipid.
