Absorption of isolated chloroplasts

To test absorption of isolated chloroplasts from spinach absorption readings were taken at a series of different wavelengths. Absorption Maxima of the isolated chloroplasts were observed at wavelengths of 440 and 680 with corresponding absorbance values of 0.792 and 0.589 were noted (Fig 1.). Absorption minima of the isolated chloroplasts was noted at a wavelength of 580 at an absorption of 0.291 (Fig 1.). This indicated that green light is reflected strongly by the spinach pigment. (Dansereau, 2013)

To test absorption of isolated chloroplasts from spinach when filters were added, filters were placed on cuvettes and time was taken in 3 minute intervals When there was no filter the absorption rose steadily throughout the twelve minutes as there was sufficient light passing through. The aluminum foil filter had very low absorption were values were as low as 0.015. The red filter had a steady increase and reached its highest point at twelve minutes and an absorption of 0.410 was noted (Fig 2). The green filter also had a steady increase in absorption throughout the twelve minutes reaching its maximum at minute twelve where an absorption reading of 0.364 was noted (Fig 2). The red filter had a steady increase in absorption and shared the same absorption value with the green filter at the nine minute mark. It reached its maxima at twelve minutes and an absorption value of 0.362 was noted (Fig 2). Blue, red and green filters had very similar absorptions throughout the experiment as shown in (Fig 2).     Lichens are a sybiotic association of fungus (mycobiont) and algae (Pediastrum boryanum . The fungus engulfs the algae, then supplies carbs, nitrogen, and vitamins while the algae photosynthesizes. This unusual relationship enables the lichen to grow in environments where neither fungus nor algae would normally be able to solely survive. They are found on every continent, ranging all the way from the freezing Arctic to the sweltering grasslands of Africa. Even though they can live in such extreme conditions, they do require clean air. This is why they are excellent environmental indicator. An indicator species is an organism whose presence or absence reflects the overall health of an environment. Lichens flourish where there is good air quality; however, they cannot survive where the air is polluted and toxins loom. This is because they have no stomata, or pores, to shut to keep the toxins out; therefore, absorbing all of the pollutants in the air. Because they are such unique organisms, they accumulate different metal anions in the air than other organisms, like plants do, for example. Lichens attract toxic metal ions. An ion is a charged atom. If the ion has a positive charge, it is a cation. An ion with a negative charge is an anion. During the ion exchange process, Hydrogen, an anion, is released and metal cations move in. This process occurs when lichens are exposed to polluted air. When the lichens are dropped into any of the metal solutions, their cell wall releases the methylene blue ions, which are replaced by metal cations. Upon their detachment, the ions integrate into the solution, which in turn, colors it blue. The darker the solution, the more metal cations taken in by the lichen.     The purpose of doing this experiment on the rate at which photosynthesis occurs, was to find out how the rate of photosynthesis could be affected by changing certain aspects of the experiment; such as the color of filter paper, which in turn changes the color of light in the experiment. My group’s hypothesis was that if the color of the filter paper was clear, then more leaves would be at surface of the cup because they had photosynthesis occurring at a faster rate. For the experiment, we deoxidized leaves by placing them in a vacuum along with a sodium bicarbonate solution. We then placed these deoxidized spinach leaves into a cup of sodium bicarbonate solution, and then placed this cup under direct light for 20 minutes to observe how many leaves floated to the top after this time period. Each cup had a different color filter paper over it to test our hypothesis; with each group in the class having a certain group of colors assigned. The average of floating disks given to us were; clear ended up with 10, purple with 6.3, blue with 8.3, and green had 2.5 of its disks floating. With these results, our hypothesis was supported with clear having the highest percentage of disks floating after 20 minutes. The rest of the data for the other color filters were provided for us, these concluded black, red, orange, and yellow filters. Yellow ended up producing a cup with the highest percentage of floating disks, which showed a similarity as it was the lightest colored filter of the four it was tested with as well.…
