To determine the effects of pH on catalase enzyme activity.Enzyme catalase Lab
Statement of the problem : Catalase breaks down H2O2 (hydrogen peroxide) into water and oxygen. H2O2 is a toxic substance formed during metabolism.
Hypothesis:
To determine the effects of pH on catalase enzyme activity and as you increase the pH the more oxygen that will be produced.
Materials:
1. 5 clean test tubes
2. HCI (acid)
3. Dilute HCI
4. Water
5. NaOH (base)
Procedure:
We start to fill test tube 1 4drops of HCI (acid), test tube 2 dilute HCI 1 drop and 3ml water, test tube 3 add 4 drops of NaOH (Base) , test tube 4 dilute NaOH 1 drop and 3ml water, test tube 5 add 3 drops of water (neutral). We filled each tube with the “liver solution to see if there is any reaction in the different test tubes.
Results :

40 mL of the enzyme solution was measured into eleven 50mL beakers that were labeled as follows: pH 2, pH
3,pH4,pH5,pH6,pH7,pH8,pH9,pH10,pH11,andpH12. 10mLofa1mol solution of each pH was added into each appropriate beaker.
A second set of eleven 50mL beakers were obtained and labeled identically to the first set. 40 mL of 1% H2O2 solution was measured into these beakers. These will be used as the reaction beakers.
Filter paper “disks” were punched out using a hole puncher. One at a time, a disk was immersed into the catalase solution for 5 seconds. The disk was then blotted on a paper towel for an additional 5 seconds. The disk was quickly immersed into the substrate solution (H2O2) and let go at the bottom of the beaker. The reaction time was measured as the time from when the disk was let go until when it floated flat on the surface of the
