Investigating the Growth of P. fluorescens Biofilms at the Air-Liquid Interface and the Variations in Biofilm Integrity
Abstract
Pseudomonas fluorescens produces biofilms in a variety of environments, and in experimental situations has been shown to develop random mutations resulting in a phonotypical change in the resulting biofilm. One particular mutation resulted in the growth of a biofilm known as the Wrinkly Spreader, which produced a thicker, stronger biofilm that had a stronger attachment to the vial. It was also more stable at the Air-Liquid interface due to its relative hydrophobicity, whereas other biofilms are kept stable by buoyancy. In the case of a floccular material growth there is moderate attachment, strength and growth but when poured onto a petri dish the biofilm breaks up into many small particles (Figure 2.). This is also a commonly found growth. A waxy aggreagate is the only biofilm from the four categories that is rarely found in experimental conditions. It is described as a strong biofilm with good growth and attachment that-when poured- breaks up into thin wafer-like fragments. The physically cohesive biofilm is the strongest biofilm of them all, and the one produced by the WspR genotype. It presents a very strong biofilm with very good growth and attachment, that when poured out onto a petri dish retains its structure as a single flexible piece. Physically cohesive biofilms also show evidence of cellulose along with other EPS substances, showing that for a PC biofilm to form it must consist of a cellulose producing miscroorganism, of which the wrinkly spreader is. (Spiers et al. 2009).
The above table was compiled after several different groups carried out the experiment, taking an observation of which biofilm type each strain was believed to be. This was done by observing the biofilms in the vials, then pouring out the biofilms into petri dishes and using the above information to sort each into its believed

    Pseudomonas aeruginosa is the bacteria of my initial unknown project. Determining which bacteria I had was completed by many steps. The one fact that I had, was that it was a gram negative bacteria because on the gram stain it appeared pink. Its cell wall is composed of a plasma membrane, periplasmic space, peptidoglycan and an outer membrane (lipopolysaccharide and protein). By looking at the agar plate it was easy to tell that it was not a swarmer and it did not have any colony pigment. It appeared as a circular, raised, undulated bacterium.     Pseudomonas tends to accumulate in biofilms in filters that are poorly maintained and in areas where pool hydraulics are poor (e.g. under moveable floors).     This was compleated by growing the bacteria on a petri dish with agar, gram staining, then examning under a microscope. The bacteria identified was M. Roseus and B. Megaterium. The identity of these bacteria was mostly determined by the positve gram stain results and the shape they where under the microscope. Both of these bacteria where identified as gram positive (as shown in the table previously). This was determined beause of the nonexistant fatty layer in the cell’s cell wall that trapped the purple crystal violate. The second big thing that identified these bacteria was the shape you could see under the microscope. While looking under the oil immersion lense, the B. Megaterium was shaped as a rod, and the M. Roseus was shaped as a circle. The diference between these two was, the rod was a much longer and skinnier shape compared to the circle whitch was smaller and more round than the rod was. The error in this experiment was that during gramstaining the second bacteria. I was able to identify three different bacteria. One was small purple circles (the M. Roseus), another was purple rods (B. Megaterium), and the last one was small pink circles that was not identified. In this lab, one of the problems that was encountered was the bacterias would not grow.     Pseudomonas putida is a gram negative and non-pathogenic bacterium with rod shape. It is usually grow in soil and water which high in oxygen content at 25-30°C.
