Lab- Dihybrid Crosses
This lab was about corn genetics- dihybrid crosses. We had to analyze the corn kernels and later on see if our hypothesis was either supported or rejected. We wanted to discover what color and coat texture was dominant (purple and wrinkled, yellow and wrinkled, purple and smooth or yellow and smooth). I had not yet done labs like these ones, but I definitely had studied about monohybrid and dihybrid, crosses. Also, I have done many activities on this aspect and other labs that were similar but not identical to this one.

To perform this lab, we had to click on the corns that were purple and smooth, yellow and smooth, purple and wrinkled or yellow and wrinkled. Then, we analyzed the data we had collected and plugged it in the Chi Square to support or reject or hypothesis. This lab was of quite simple procedure and did not require many steps.

My calculations were my Chi-Square tests, which I am posting here under the discussion box.

In this lab, I counted the kernels in corn based in color and coat texture. In the hybrid crosses the purple and smooth showed up the most (later on we can see these are the dominant alleles when we plug these values in the Chi-Square test table). If we were to do Punnett Squares (parents of hybrid crosses and two heterozygous parents) to discover the estimation of the outcomes (the percentage of chance the outcome will be a certain thing), we would see that expectations are that purple and smooth alleles will cover 9/16 of the corn and that there is 9/16 probability that the kernel will end up being purple and smooth. With the Punnett Square, we can see that purple and wrinkled alleles have 3/16 chance of occurring, just as yellow and smooth alleles. The yellow and wrinkled alleles are 1/16 probable to occur. With these numbers we can come up with the expected ratio for this cross, which is 9:3:3:1.     To begin the fast plants experiment, each group received the F1 generation seeds for their designated phenotypes. After receiving the seeds, the seeds were then planted into Styrofoam “quads” allowing the seeds to germinate. The seeds were then allowed to grow under a fluorescent light bank for 5 weeks. Throughout the growing of the seeds, each plant was continuously watered and adjusted the light bank to help the plant grow more efficiently. After letting the seeds germinate for approximately 14 days, after two or three flowers had opened, a bee stick was used to transfer pollen from one plant to another. Each group then used the bee stick again 2 and 4 days after the initial pollination. Once all of the transferring was complete, each unopened bud on the plants were pinched off. Approximately 21 days after pollination, the seeds were ready to be harvested. Each quad was removed from the watering tray and allowed to dry for 5 days. After the plants were dry, each group removed the dry seedpods from the plant. We then removed the seeds from the pod. To start the germination process, a moistened piece of filter paper was placed into a petri dish. 40 seeds were neatly placed into each petri dish until there were no seeds remaining. Once all the seeds were placed in the petri dish, the petri dishes were placed in a plastic bag and set to germinate for approximately 48 to 96 hours in the window at room temperatures.…

    Statistics is an area that most A-level biology students (and their teachers!) find difficult. The formulae are often complicated, the calculations tedious, degrees of freedom mysterious, and probability tables confusing. But in fact students need no longer grapple with any of these. In real life, biologists and statisticians rarely use calculation and tables these days, but instead use statistical packages such as Minitab or SPSS. But it isn’t even necessary to buy an expensive statistics package, since spreadsheet software such as Excel has most of the common statistical tests built-in. When using statistics, the first hurdle is to decide which statistical test to use. Figure 1 (overleaf) is a flow chart showing when to use the various tests described in this article. There are many other possible statistical tests, but this flow chart should be more than sufficient for A-level biology students. It briefly summarises the Excel formulae and how to interpret the results, so it can be used as a handy guide on its own once the student is familiar with the tests. This flow chart should be used when designing an experiment, not after the experiment is complete. This will ensure that the correct kind of data are collected so that the statistical test will be valid.
