Experiment 1: Carbohydrates
Separation Techniques Based on Molecular Size

Purpose The purpose of this experiment is essentially two-fold. Two of the four experimental procedures of this laboratory will be used to gain a conceptual and practical understanding of separation techniques used to isolate monomers from their respective polymers.
Namely, the techniques that will be explored are dialysis and gel filtration chromatography.
In the remaining two experimental procedures, colorimetric assays will be used to detect the presence of certain carbohydrates. Glucose oxidase and Iodine Reactions will be performed in conjunction to demonstrate the procedure of such assays.

A cellophane dialysis tube (1.6 cm diameter, 2.0 mL/cm, MW cut-off:12000-14000) was clipped on both ends and filled with 10 mL of a starch, glucose solution after being soaked in water for 5 minutes to remove any storage additives. The dialysis tube was placed within a 200mL beaker filled with distilled water and paced on a stir plate. A wavelength of 420 nm was chosen while recording the absorbance intensity of glucose, while a wavelength of 600 nm was chosen for starch as the respective molecules absorb most efficiently at these wavelengths. C1 is the concentration of glucose or starch in the internal solution and C2 is the concentration of glucose or starch in the external solution.

    Spectroscopy is the study of light. A spectrophotometer is a machine used to determine the absorbance of light at any given wavelength. It does this by using a source of white light through a prism, which gives multiple wavelengths that can be individually focused (Ayyagari and Nigam, 2007). Substances are put into cuvettes that are glass or quartz containers that light can easily travel through. The light that is being focused travels through the substance gets absorbed by the substance and is reflected back and read by galvanometer which had the ability to detect electric currents (Verma, R). The absorbance reading is then given, absorbance is usually between 0.0 – 2.0, any higher than 2.0 may mean not enough light is getting through to the galvanometer (Bhowmik and Bose, 2011).
