ABSTRACT
In this experiment, the cell growth of yeast is measured by using spectrophotometer and hemocytometer. We learnt how specthophotometer and hemocytometer use and also we learnt qualifications of hemocytometer and spectrophotometer.Serial dilution was used for this experiment and it was very important.Because of the serial dilution,we measured the number of yeast cells. The graph of growth curve was drawn and bacterial life cycle was understood with the graph. The purpose of the experiment was to calculate and draw bacterial growth curve.
INTRODUCTION
In this experiment,we learnt cell growth.Cell growth means increase in the quantity of cellular constituents.Cells in culture double its number because they produce two daughter cells by dividing.Generation time is the constant time when cells in the culture double their number. This kind of growth depends on some requirements such as type of the cell and environment. Two different parameters can be used to calculate growth for unicellular organisms such as bacteria and yeast:changes in cell mass and cell number. There are some different ways for measuring the cell growth with cell mass.Physical measurement:dry weigth, wet weigth or volume after centrifuge and: Chemical measurement chemical components such as N,total protein and DNA and Chemical activity:rate of , CO2 production or consumption; Turbidity (spectrophotometric) measurement: based on turbidity and measure both dead and alive cells.Spectrophotometer can measure intensity of the light source wavelength.Spectrophotometers are important in terms of spectral bandwith and linear range of absorption or reflectance measurement.Spectrophotometers are used for physics, material science, chemistry, biochemistry and molecular biology experiments. Why must spectrophotometerbe set to

Yeast are eukaryotic species and make up approximately one percent of species in the kingdom Fungi. One of the most well studied yeast species, Saccharomyces cerevisiae, commonly known as bakers yeast, and also used in the fermentation of alcohols, is a model species for the study of eukaryotic cells. They are known to reproduce either sexually, by mitosis, or asexually, by budding, depending on environmental conditions. Yeast grow and reproduce by producing enzymes to catabolise polysaccharides. Upon maturation the parent cell passes its genetic information to the bud which then becomes a genetic copy of the original cell and adds to the population. A growth curve demonstrates the four stages of population growth for a yeast cell as follows:…

Yeast is a very important group of fungi. The common yeast used in baking bread grows very fast. The basic idea in this lab is that the yeast will use an energy source and in doing so, will produce carbon dioxide gas. You’ll measure the amount of carbon dioxide (CO2) released during the growth of yeast. The growth of the yeast stops when the energy source has been used up by the yeast and CO2 production slows down or stops.…

Abstract. -- The main focus in this experiment was to find out if antibiotics affect yeast cells culture when added. The observation of the two cultures along with the yield of yeast cell reproduction between the two cultures was the sole purpose of the experiment; culture-A (- antibiotic) and culture-B (+ antibiotic). Our hypothesis stated whether antibiotic effects yeast cells cultures. This theory was tested by having a chamber with quadrants that contain the culture, a hemacytometer, was used to carry out this experiment. After the data was collected from using the hemacytometer, the results of the difference between the two cultures was to be determined through a t-test. As the test was conducted, the study shows that the difference between lacking an antibiotic and containing one is not different. An antibiotic in culture-A and culture-B does not affect the number of yielded yeast cells.…

In multicellular organisms (commonly Eukaryotes), creating more cells during cell division is the cause of growth. When it is certain that the daughter cells will have everything they need to survive and thrive, Mitosis begins, starting the process of Cell Division. How large or small the cell's surface area is determines the amount of material entering and leaving the cell. Also, the amount of space in the cell?cell's volume determines the rate that those same materials are used . There are many things that can affect cell growth. For example, an abundance of food and a constant temperature can increase cell growth while the opposite can decrease cell growth.…
