Gatorade Spectrophotometer
The purpose of this lab was to use a spectrophotometer to find the concentrations of glucose in varying samples of Gatorade. The varying executions included changing the temperature of reaction (room temperature instead of 40 degrees centigrade), changing the time for reaction (20 minutes in the water instead of 45 minutes), and changing the amount of enzyme added (0.1mL of enzyme. The results in from this lab gave data points with respect to ferricyanide. Fortunately, ferricyancide and glucose have a 1:1 ratio meaning that the amount of glucose can be deduced. For the first 7 samples required the spectrophotometer to determine the absorptivity, which was run at 420nm. These data points could then be used to find a line of best fit for the later calculations. The R^2 value was given at 0.986. This was after deleting a value from the data set because it was pulling the R^2 value down. The line of best …show more content…
One source of error came from washing out the test tube with the next sample before running the test. This means that there could have been remnants of the old sample in the new sample. Another source of error could have been from lack of precise timing causing the reaction to not go as long or longer than desired. Of course, each solution was made by hand leaving room for human error. These errors include over injecting of solution.

Conclusion
This experiment found the amount of ferricyanide which was then used to find the amount of glucose thanks to a 1:1 ratio. This could then be used to find the concentration of glucose in various samples of Gatorade (15 solutions). Then the lab was split altering 3 different variables (Time, Temperature, Enzyme amount). This illustrated the variable that had the biggest impact on the results.

1. No, it would not work because sucrose is a polysaccharides but this enzyme interacts with monosaccharides. This could only work if they were broken down or if a different enzyme was

    Sucrose was successfully transformed into D-glucose and D- fructose (disaccharide to monosaccharides) through acid-catalyzed hydrolysis. This is obvious when comparing the experimental specific rotations of sucrose and invert sugar with the theoretical values of sucrose (+66.5°) and invert sugar (-39°). However, my partner and I experienced a small complication with the addition of the acid, HCl. After adding the required amount of HCl to the mixture, we experienced no color change for about an hour. Due to this problem, we came up with a solution to add more HCl due to the possibility that the acid was absent from the reaction. An additional 16 drops of HCl was added during the heat reflux phase. The solution displayed a tint of yellow after about 5 minutes of refluxing, and then it was completely yellow after another 5 minutes.
