ABSTRACT
High Performance Liquid Chromatography has been used to evolve an analytical procedure for the evaluation of the content of paracetamol in the bulk, dosage forms and in urine, a body fluid. Separation and resolution have been achieved with a combination of methanol and 2.5% acetic acid (15:85) on a reversed-phase column at ambient temperature. Elution was isocratic with UV detection at 257nm.
Internal standard calibration method was used for quantitation with caffeine as the internal standard.
Mean retention times for paracetamol and caffeine were respectively 2.61 ± 0.13 min and 11.98 ± 0.72 min . The calibration curve was linear over the range 0.1-5.0μg/ml. The method was also suitable for the assay of paracetamol-codeine combination drug as well as estimation of the amount of constituents in urine when the wavelength of UV detection was 245 nm with acetanilide as the internal standard.
Keywords: Chromatography, isocratic, internal standard, in vivo and in vitro
INTRODUCTION
Paracetamol (N-(4-hydroxyphenyl) acetamide) tablets are listed among the essential drugs selected for the health care delivery system in Ghana.

Paracetamol is very much used for antipyresis and analgesia without prescription. The drug is useful in mild to moderate pain such as headache, myalgia and postpartum pain. It is a very good alternative for mild to moderate pain in patients who cannot take aspirin because of allergy, haemophilia, history of peptic ulcer and asthma. (Katzung, 1989).
As a result of the Ghana government’s policy of generic prescribing, the

The second experiment of the semester focuses on separating and identifying the components of Panacetin, which is a common medication that relieves pain. The label reveals that Panacetin is composed of sucrose (10%), aspirin (40%), and Tylenol (50%). Although this information is reported, the true composition of Panacetin is questionable. While sucrose and aspirin are present in Panacetin, Tylenol may be replaced by an unknown component (acetanilide or phenacetin). Separation of the components of a sample of Panacetin may prove valuable in identification of the unknown component, as well as the true percent composition of each component of Panacetin.…

To be able to understand how organic soluble compounds could be isolated from non-organic components, a chromatographic separation process was used. The objective of the analgesic drug identification is to isolate the organic compounds/ active ingredients in an unknown drug. The identity of the drug was determined through melting point, percent recover, and Rf values. The comparable data for acetaminophen from the manufacturer is 500mg of active ingredient. After conducting the experiment-extraction, distillation, and evaporation- 451mg of active ingredient. This relates to a 68.1% recovery. The melting point was between 158 and 165 degrees Celsius, which is almost ten degrees lower than the published melting point of acetaminophen. After conducting a TLC plate, the Rf value was calculated to be .4268, the same as the standard Rf.…

Generally, calcium antagonists are important in the treatment of hypertension. Among all known hypertensive agents dihydropyridine derivatives (DHP) have gained the most approval. Therefore, selective, sensitive analytical methods for the quantitative evaluation of the drugs and their metabolites are important for the successful conduct of preclinical and/or bio-pharmaceutical and clinical pharmacology studies. Bio-analytical method validation for paracetamol drug includes the parameters such as accuracy, precision, selectivity, sensitivity, reproducibility and sensitivity. All this validation methods are applicable to bio-analytical methods such as Gas Chromatography and High Performance Liquid Chromatography. Unfortunately the most undesirable feature of drugs containing the DHP (dihydropyridine) group is high sensitivity to light .Such drugs are regarded as photo labile. Therefore the photo stability testing has become an important stage of new drug design, production and…

Aspirin is used for a wide range of maladies beginning at headaches and ranging from menstrual pains to use in preventing cancerous tumors. Aspirin is used antiplatelet. This means that it is used to stop platelets from congregating and forming blood clots. This helps to prevent heart attack and strokes. Aspirin is a non-steroidal anti-inflammatory drug (NSAID). NSAIDs are medications with analgesic, antipyretic and in higher doses anti-inflammatory effects. An analgesic is a drug that produces temporary pain relief without inducing a stupor and has no drowsy-side effects. Aspirin is said to be antipyretic because it can be used to reduce or to cure fevers and its anti-inflammatory properties allow it to be used to reduce swellings and other bruises (inflammations) in and outside of the body. Non-steroidal means they are not steroids, which often have similar effects.…

In analyzing the chromatography of analgesics, thin layer chromatography (TLC) was used. A very thin (micron) film of silica is coated on a glass plate or a sheet of aluminum or plastic. The ingredients of commercial analgesics were characterized in comparison with chromatograms of aspirin, Acetaminophen, caffeine and mefenamic acid. After the TLC was placed inside a UV chromatography box, it appears that the sample analgesic contains D, which is mefenamic acid.…
