A precise, accurate and rapid RP-HPLC method has been developed and validated for the simultaneous estimation of Tamsulosin Hydrochloride and Finasteride in tablet dosage form. Chromatography was carried on RP – 18e Hiber RT (250 × 4.6) column using Water:Methanol (30:70%) v/v as mobile phase at a flow rate 0.7 ml/min and the effluent was monitored at 225 nm. The retention times of Tamsulosin Hydrochloride and Finasteride were 5.4 and 15.4 minutes respectively. The limit of detection for Tamsulosin Hydrochloride and Finasteride were found to be 15 and 50 ng/ml respectively and their limit of quantification were found to be 100 and 1250 ng/ml respectively. Tamulosin Hydrochloride and Finasteride were found to be linear in the range of 0.3 to 10 µg/ml and 3.75 to 125 µg/ml respectively. The developed method was validated in terms of linearity, accuracy, precision, specificity, limit of detection, and limit of quantification as per ICH guidelines. System suitability parameters like number of theoretical plates (N), peak asymmetry factor (As), and resolution (Rs), etc. were studied and it was found that all results were within limits. The proposed method was successfully applied for simultaneous determination of drugs in tablets.

Immediately, 100 µL of BEC-HRP conjugate diluted using PBS (1:15000) was added to the standard solution. The microtiter plate was incubated for another hour while shaking at 750 rpm. Consequently, another three cycled washing step was performed, 200 µL of the freshly prepared HRP substrate solution was added to each well. The HRP substrate solution25 was 21 ml citrate buffer (220 mM sodium citrate monobasic, pH 4.0) + 8.1µL H2O2 (30%) + 525 µLTMB solution (40 mM TMB, 8 mMtetrabutylammonium borohydride, in N,N-Dimethylacetamide). The microtiter plate was incubated for 30 minutes. Eventually, the color development was stopped by adding 100µL of 1 M H2SO4. And therefore, the absorbance was measured using plate spectrophotometer SpectraMax Plus384 (Molecular Devices, Ismaning, Germany) at 450 nm with 620 nm as a reference. Four parameter fit (4PL) was used for measuring Cocaine ELISA calibration curve; A (upper asymptote), B (slope at the inflection point), C (the inflection point on the calibration curve), and D (lower…

Methodology: Using auto-pipette (20-100 microlitre), pipetted 0.06 mg of Taxifolin was added to 10ml conical flask and made up to the mark (10ml) with H2O.This procedure was repeated with the same amounts foe silymarin stock solution and silybum marianum extract. Taxifolin was transferred to 2ml brown bottle for HPLC auto sampler, this was also done to silymarin stock solution. Silybum marianum extract was filtered via a syringe filter (0.2 microlitre). The three samples were tested for concentrations of taxifolin via HPLC.This process was repeated a further 20 times with different amounts of taxifolin, silymarin stock solution and silybum marianum extract. Results of Taxifolin standard shown in table 1 and figure 1.…

The drug components were distilled in a test tube and were used to spot the TLC plate. The spot were about 1 cm between on another to give enough room for distinctive separation once all the chemicals were spotted on the TLC plate, it was inserted into the developing chamber and observed carefully we removed it after a minute or so and let it dry and then placed it in the short UV light. And the spots were outlined by the pencil and the areas were dotted with the greatest density this then helped us calculate the value of Rf the value of our unknown was around 0.23 and 0.25. And by that what we hypothesized were correct which was s4- Acetaminophen and the Rf supports that…

The majority of medications currently developed and prescribed are in the form of tablets or capsules due to their ease of consumption by the patient. The identification and characterisation of tablets and capsules is a vital process within the drug development timeline, as the pharmaceutical form of a substance can impact significantly its stability and bioavailability [23]. Taylor & Langkildl explain how useful it could be to be able to identify these solid-state medications in situ, particularly where different salts are present [24]. Tablets often contain low doses of API, which can present analytical…

Generally, calcium antagonists are important in the treatment of hypertension. Among all known hypertensive agents dihydropyridine derivatives (DHP) have gained the most approval. Therefore, selective, sensitive analytical methods for the quantitative evaluation of the drugs and their metabolites are important for the successful conduct of preclinical and/or bio-pharmaceutical and clinical pharmacology studies. Bio-analytical method validation for paracetamol drug includes the parameters such as accuracy, precision, selectivity, sensitivity, reproducibility and sensitivity. All this validation methods are applicable to bio-analytical methods such as Gas Chromatography and High Performance Liquid Chromatography. Unfortunately the most undesirable feature of drugs containing the DHP (dihydropyridine) group is high sensitivity to light .Such drugs are regarded as photo labile. Therefore the photo stability testing has become an important stage of new drug design, production and…

Chromatograph method is a method of separating mixtures of two or more compounds. Two phases are important in this method; one that is stationary and one that is moving. Chromatography works on the principle that different compounds will have different solubilities and adsorption to the two different phases between which are to be partitioned. Thin Layer Chromatography (TLC) is a solid-liquid technique. The mixture is observed when it is in two different phase; a solid (stationary phase) and a liquid (moving phase). Thin Layer Chromatography (TLC) is a technique which is sensitive, cheap and fast. A thin layer chromatograph plate is used to identify drug component under UV. It is used to support the identity of a compound in a mixture when the Rf of a compound is compared with the Rf of a known compound.…
