Chem 201

10/02/2012

Objectives :

1)To purify samples of organic compounds that are solids at room temperature.
2)To dissociate the impure sample in the minimum amount of an appropriate hot solvent

Chemical Index : Naphthalene; mp 82 *C 1,4-Dichlorobenzene; mp 56 *C

4-Chloroaniline; mp 68 - 71 *C; pKb 4.15 Ethyl 4-Aminobenzoate; mp 90 *C; pKb 4.92
Procedure :

Partition Coefficient of Benzoic Acid
1) Carefully weigh approximately 100 mg of benzoic acid and place it into the centrifuge tube (or test tube). The dichloromethane addition should be carried out in the hood. Using a 10-mL graduated cylinder, add 2 mL of dichloromethane to the centrifuge tube. Mix until the benzoic acid dissolves.

2) Add 2 mL of water to the benzoic acid dissolved in dichloromethane. Cap the centrifuge tube and shake (or use a Vortex mixer) until the two phases have been thoroughly mixed. Vent the vial (by releasing the cap-seal) and then allow the two layers to separate.

3) With a disposable pipet , carefully remove the entire mixture. Allow the two layers to separate in the pipet. Drain the lower layer dichloromethane from the pipet into a test tube. Discard the pipet containing water. Add a rice sized portion of anhydrous sodium sulfate and shake the mixture. With a new disposable pipet transfer the CH2Cl2 layer into a large pre-weight test tube.

4) Under a hood, evaporate the CH2Cl2 layer by warming the test-tube in a water bath (CAUTION: This evaporation must be done in a fume hood). After the solvent has been completely removed, finalize the drying of the benzoic acid by placing the test tube under vacuum and pre-weigh the dry test-tube containing the residual benzoic acid.

5) Calculate the dichloromethane/water partition coefficient for benzoic acid.

First place a small amount of the spinach provided in the mortar, just enough to cover its bottom; combined with the sand provided to break down the call walls. A 1:1 mixture of hexane and acetone was determined to best solvent for this extraction. Knowing this add one mL of the pure hexane and one mL of the pure acetone to the mortar. Grind the contents until the cell walls have been adequately broken. Transfer the mixture to a centrifuge tube to separate the sample into layers, to help remove the water. Once the sample is finished spinning extract the organic layer and place in a test tube.…

Add 8.0 mL of dichloromethane to the 0.40 g of Panacretin and shake thoroughly, then place in a centrifuge, at three (3) speed for three (3) minutes. Extract the Sucrose and weigh using a digital scale (must be tared for accuracy). Separate the 8.0 mL of DCM to two (2) test tubes, each containing 4.0 mL of DCM. Add 4.0 mL of NaHCO3 (Sodium bicarbonate) to each of the two (2) test tubes and stir using a stirring rod until two (2) layers are visible, a thin top layer and thick-filmy bottom layer. Extract the bottom layers from the two (2) test tubes to a vial using a glass micropipette and a rubber suction cap/top and seal immediately. Conjoin the two (2) top layers to a 50 mL flask and add 1.0 mL of 6.0 M HCl to the beaker containing the solution and stir until the fizzing subsides. Test the pH of the solution with 6.0 M HCl added using pH paper, which has a color chart to analyze the results. Continue to add 1.0 mL of 6.0 M HCl until the pH of the solution is two (2) or less. Place the solution with the pH of 2 or less into an ice bath/cold water bath to form crystallization of aspirin for approximately five (5) minutes. After the five (5) minutes, establish a funnel system using a Hirsche Funnel attached to a vacuum filtration system located on/in the hood, to extract the Aspirin crystals formed from the ice bath/cold water bath. Weigh the crystals of aspirin using a digital…
