BIOL160: Lab Report 1
January 30, 2013

Lab partners: Park Jin Mun 0304299 Tan Zhi Chen 0306521

Hypothesis

1) If a calculated absorbance level is directly related to the concentration of albumin, then a higher concentration of albumin will lead to higher absorbance level.

Results

Table No. | Albumin(cm3) | KCl (mL) | Biuret Reagent (mL) | Absorbance | 1 | 0 | 5 | 3 | 0.049 | 2 | 1 | 4 | 3 | 0.112 | 3 | 2 | 3 | 3 | 0.258 | 4 | 4 | 1 | 3 | 0.39 | 5 | 5 | 0 | 3 | 0.418 | 6 | 5 (A) | 0 | 3 | 0.198 | 7 | 5 (B) | 0 | 3 | 0.347 |
Table 1) Test Result

Table 1 is the results of seven test tubes with a different amount of solutions. 3 mL of Biuret Reagent was added to all seven test tubes and absorbance was measured with spectrophotometer. It is observed that the absorbance level increases as the amount of albumin is added more in different test tubes, while the amount of KCl decreases.

Concentration of albumin in mg(x-axis) | Absorbance (y-axis) | 0 | 0.049 | 2.5 | 0.112 | 5 | 0.258 | 10 | 0.39 | 12.5 | 0.418 | (A) | 0.198 | (B) | 0.347 |
Table 2) Analyze of two factors, concentration of albumin and absorbance The concentration of albumin in the standard albumin is 2.5 mg/cm3of 0.5 M KCl. Hence 1.0 cm3of albumin is calculated to 2.5 mg of the concentration of albumin. For 6th and 7th test tube were unknown labeled as (A) and (B).

In order to create a standard curve we created 5 dilutions of solutions containing red dye #40. Concentrations of .666 ug/mL, 1.667ug/mL, 3.333ug/mL, 13.333ug/mL, and 20.000 were measured in a spectrophotometer set to a wavelength of 504 nm to find the amount of light they absorbed. Using Microsoft Excel, we entered the absorbance values as the Y coordinates and the concentrations as the X coordinates to make a line of best fit and that was our standard curve.…

For the first part of the experiment, 4.0 mL of the 0.2 M NaOH solution were poured into a small test tube. The test tube was wiped and placed into the sample holder of the spectrophotometer and the transmittance was set to 100.0 for calibration. The test tube was removed from the sample holder and 1 drop of phenolphthalein was added and inverted several times to mix. With the Spec-20 set to absorbance mode, the test tube was quickly placed into the sample holder and the absorbance was recorded every 5 seconds for 360 seconds. For the second part of the experiment, the same calibration steps were repeated and then, 2.0 mL of the 0.2 M NaOH and 2.0 mL of the 0.2 M NaCl were added to a small test tube. After this step, 1 drop of phenolphthalein was added and inverted several times to mix. With the Spec-20 set to absorbance mode, the test tube was quickly placed into the sample holder and the absorbance was recorded every 5 seconds for 360 seconds. After each part of the experiment, the test tube was removed from the sample holder and its contents were placed in the waste.…

The results of our experiment showed the solutions in both tube 1 and tube 2 increasing in absorbency in the first eight minutes but then tube 1 continued to increase while tube 2 began to balance out. Tube 3, our blank, managed to stay at 0nm the entire twenty minutes. From this data, we can conclude that our hypothesis was supported that EDTA had a greater change in absorption over PTU.…
