A Factor That Will Affect
Enzyme Activity

The Question
Effects on Enzyme activity: investigating the effects of mixing Hydrogen Peroxide with Catalase extracted from potatoes at different temperatures, and then comparing the results with the same method, except to replace the Potato juice with MnO2

Hypothesis
Since enzymes are a type of protein, they will be destroyed at high temperatures and activity rates will go down at low temperatures. Also, if done at low temperature and then heated to room temperature, the enzyme may thaw out and start reacting again.

Variables Variables | Units | Range | Independent | temperature | Celcius | 0, 21, 50, 80 | Dependant | Potato juice and MnO2 | Centimeters | 0~5 |

Controlled variable | Units | Effects | Control By | Amount of H2O2 | mL | Amount of bubbles | Using syringe |

Equipment
Syringe
Beaker
Ice
Hot Plate (Heating Pa)
Test tubes
Potato
PH7 soap
Thermometer
H2O2
MnO2 (non-biological catalyst)

Method 1. Prepare the potato juice by grinding or squeezing and pour it into a beaker. 2. Then pour a little of the soap in. This is to help last the bubbles so they do not pop quickly. 3. Prepare each test tube, five of them, by each pouring in 5mL of potato juice. 4. Prepare the stages for each test tube. i. Put ice in a beaker with water and put in the test tube ii. Leave it in a test tube holder at room temperature iii. Turn on the heating pan and put the beaker on top of it. After the water has been heated enough, put in the test tube. 5. After it has all been set, put in the H2O2 in every test tube set. 6. After doing steps 1~5 replace the potato juice with MnO2 (0.1g) and do the experiment over.

Potato | Description of Observation | 0 | Very limited reaction could be seen. There were only a small amount of bubbles being formed. Reactions took a while after dropping in the H2O2. | 0 and then room temp. | After taking the test
