Figure 1: Schematic illustration of the principle of Biacore SPR to measure the interaction between analyte molecules in the flow with immobilized receptor binding sites on the surface. A) SPR uses an optical method to measure changes in the refractive index near the surface of a thin gold film on a glass slide. At a critical angle of incident light on the surface-solution interface an electromagnetic field or evanescent wave is generated. This is detected as absorption of light and a decrease in the intensity of the reflected light. The generated evanescent wave extends a short distance (≈ 300 nm) into the solution and is sensitive to changes in the medium. To overcome this, a very common strategy is to use secondary molecules such as antibodies, to capture the target protein on the surface. An antibody that recognizes the membrane protein of interest can be immobilized covalently on the surface and subsequently capture the target protein when it is injected. Opposite to covalent immobilization, where proteins are immobilized randomly, the availability of monoclonal antibodies directed against specific epitopes on the target protein makes it possible to immobilize proteins in controlled orientations …show more content…
In addition to stabilize the biological function of captured membrane proteins, nanodiscs have some benefits when used together with SPR. Nanodiscs provide access to both sides of the lipid bilayers. Thereby it is possible to avoid problems with orientation and access to integral membrane proteins that can occur when lipid vesicles or supported membrane mono- or bilayers are used to capture membrane proteins on the sensor surface. In addition, the MSP encircling the nanodisc lipidbilayer can increase the flexibility of immobilization strategies; the MSP can be genetically engineered with various affinity tags useful for immobilization on a complementary antibody sensor surface.     Before arrival, the COS-7 cells were grown in culture dishes submerged in a growth medium that contained bovine serum. Two coverslips covered in the COS-7 cells were provided. After washing the coverslip with Phosphate Buffer Saline (PBS), one coverslip was treated with the control solution, and the other was treated with the X drug. Then, in order to preserve the cell, methanol was added to both coverslips. Since methanol coagulates proteins, it was also used to permeabilize the cell membrane. Once the cells were fixed and permeabilized, blocking buffer was added to each coverslip to block the nonspecific antibodies in order to keep them from binding to everything in the cell. Then the primary mouse antibodies were added to the cells, and the secondary antibodies are added to bind to the primary antibodies.     When a B cell recognizes an antigen, the B cell has antigen receptors with 2 heavy chains and two identical chains joined by disulfide bonds. This gives a Y shape fore each antigen receptor protein on a B cell. An antigen receptor protein consists of two sites: a constant region (C) and a variable (V) region. In the constant region, the amino acid sequences remain similar to another antigen receptor from another B cell. What determines the main difference in targeting specific types pathogen for each B cell is the variable region. A variable region of an antigen receptor from one B cell has completely different amino acid sequencing than the variable region of another antigen receptor from another B cell. Thus, the variable region forms a bond with the specific antigen. This in terms activates the B cell to secrete antibodies. Similar to the Y-shaped structure of the antigen receptor, antibodies target on specific antigen binding site on a pathogen. Such process inactivates the infections from pathogen. After all, the variable region of an antigen receptor of a B cell provides a diverse variety of combinations of amino acids. The present agreed on model, created in 1972 by S. J. Singer and G. Nicolson, is called the fluid mosaic model. This model depicts that proteins (integral and peripheral) form a mosaic since they are floating in a fluid layer of phospholipids, which makes up the components of the cell membrane (along with cholesterol).     All living cells have a cell membrane and it is probably the most important organelle of a cell. The cell membrane is composed of a phospholipid, proteins and carbohydrates. Membranes can contain phospholipids with different fatty acids which affects the strength and flexibility of the membrane. There are two different proteins in cell membranes.
