ANTIMICROBIAL ACTIVITY OF PROPOLIS COLLECTED IN DIFFERENT
REGIONS OF SULAIMANI PROVINCE-KURDISTAN REGION/IRAQ

ABSTRACT
The aim of this study was to investigate the activity of (28%) ethanolic extract propolis (EEP) of [Bazian, Pshdar, Sharbazher,
Khormal, Kanakawa and Sulaimani (city center)] against (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli,
Pseudomonas aerugenosa, Klebsiella pneumoniae, Proteus mirabilis and yeast Candida albicans). Two methods were employed for propolis activity evaluation; firstly by agar well diffusion method using 40µl of propolis extracts and ethanol 70% as the solvent control per well against tested bacteria and fungus and more commonly used antibiotics were used in comparison to propolis samples. The second method was employed by measuring Minimal Inhibitory Concentration & Minimal Bactericidal; Fungicidal
Concentrations (MIC, MBC & MFC) using two fold dilution method for propolis extracts and ethanol 70% as a solvent control in serial concentrations. The Results exhibited that propolis is more efficient than some antibiotic discs. The most sensitive bacteria against (EEP) with mean inhibitory diameters (22-26) mm was S. epidermidis followed by S. aerues and C. albicans (15-22) mm.
Tested Gram negative bacteria have lowest inhibitory zones (7-9) mm. Results of MIC & MBC showed S. aureus with (0.1750.7mg/ml) followed by S. epidermidis & C. albicans with (0.7-1.4mg/ml). Among tested Gram negative bacteria, all (EEP) samples showed relatively strong inhibitory action on P. mirabilis (14mg/ml) and weak inhibitory action on P. aerugenosa & K. pneumoniae

    To study whether a microbial product qualifies as an effective antibiotic, a standard procedure called the Kirby-Bauer method is employed. This method, which is the procedure recommended by the US Food and Drug Administration, was devised by William Kirby and A.W. Bauer in 1966. In the current protocols involved in the Kirby-Bauer method, Mueller-Hinton standard agar is used as the medium for bacterial culture. The pH of the standard agar is 7.2 to 7.4 and it is poured exclusively to a depth of 4 mm. The medium is heavily inoculated with bacteria and paper disks containing enough of the antibiotic under study to create an optical density of 1 (the McFarland standard) are placed on top of the cultures. By examining the results of incubation in the form of a zone of inhibition around each disk after incubation, it can be determined how effective each antibiotic is against any given bacterium. A minimum inhibitory concentration can then be deduced for the given antibiotic vs. the specific bacterium tested so that appropriate dosage may be determined.     The purpose of this lab was to determine the effectiveness of antiseptics, disinfectants, and antibiotics on bacteria. The hypothesis was that if bleach was used, it would be the most effective because bleach is commonly used to clean and disinfect various things. The variables that were tested were antibacterial soap and Scope mouthwash for the antiseptics; bleach and ammonia for the disinfectant; and Cipro, erythromycin, and tetracycline for the antibiotics. All of these chemicals were used on the bacteria M. luteus. Two Petri dishes were covered in the bacteria and split into four quadrants, in which each had a disc containing one of the chemicals stated above. One quadrant was left alone with no chemicals for the control group. The dishes were then left for the bacteria to grow, and once obtained again it was obvious that some of the bacteria was killed by the chemicals. In individual data, there was a zone of inhibition of 3mm in the antibacterial soap; 10mm in the Scope mouthwash; 2mm in the bleach; no zone of inhibition around the ammonia; 10mm in both the erythromycin and the tetracycline, and 15mm for the Cipro. The average length of the halo of inhibition in antiseptics was 8 mm in the E. coli, and 6 mm in the M. luteus. The average length of the halo of inhibition in the disinfectants was 12mm and 11mm respectively. For the antibiotics, it was 7mm and 9mm respectively. The data represented the hypothesis because for both the E. coli and the M. luteus, the largest zone of inhibition was in the disinfectant; and more specifically, the averages were 28mm in the E. coli and 18mm in the M. luteus in the bleach. In some Petri dishes, the bleach also killed bacteria in the other quadrants, indicating that it killed a lot of bacteria. This also affected some measurements for the other chemicals, because the zone of inhibition for the other chemicals around the bleach could have been caused by the bleach instead of the other chemical.…

    In this lab, chemotherapeutic agents were evaluated by the disk-diffusion method. Chemotherapeutic agents are placed on the surface of a Petri plate containing Mueller-Hinton agar, which allows the agents to diffuse freely, and a growth medium is incubated over its’ surface. During incubation, the chemotherapeutic agents move from levels of high concentration, to low levels of concentration.     The antibiotic that was the most effective was Streptomycin. This was due to Streptomycin being an antibiotic that works on gram negative bacteria. Gram-negative bacteria are bacteria that do not retain crystal violet dye in the Gram staining protocol whereas Gram-positive bacteria are those that are stained dark blue or violet by Gram staining.
